The Role of Serotonin

The neurotransmitter serotonin has a well-described role in regulating mood states and cognition in the human brain. Insufficient serotonin activity is centrally related to the hypotheses of mood imbalance[source]. Historical use of pharmacological agents, such as the serotonin selective reuptake inhibitors (SSRIs) approved in the late 1970s, showed the effectiveness of elevating serotonin in the central nervous system. A recent study correlated positive and negative mood states to the relative amount of radiolabeled C11 5-HTP[source], highlighting the significance of 5-HTP to serotonin conversion in the regulation of emotion (Figure 1).
Alhough pharmacological agents targeting serotonin are effective at elevating mood, the multitude of side effects has prompted alternative methods of increasing serotonin levels in the brain. Since the 1980s, there has been considerable focus on serotonin precursors mitigating poor mood. Several clinical trials using the serotonin precursors 5-hydroxytrypophan (5-HTP) and tryptophan have produced strong evidence for the use of these bioactive compounds in elevating mood[source]. The inclusion of 5-HTP and tryptophan, combined with the co-factor vitamin B6 in the New MOOD™ formula, can help the body enhance the active production of serotonin within neuronal cells.

Serotonin Precursors Elevate Mood

Cells outside of the brain also produce and utilize serotonin, yet this peripheral serotonin cannot be used as neurotransmitter because of its inability to cross the blood-brain barrier. Instead L-tryptophan is ingested through diet and is enzymatically converted to 5-HTP with the help of vitamin B6. The synthesis of serotonin is regulated by the rate-limiting step of L-tryptophan to 5-HTP. In other words, supplementing the body with 5-HTP allows for a rapid conversion to serotonin production. Meanwhile, the inclusion of L-tryptophan permits a slower, more prolonged production of serotonin.
Several studies have tested the effectiveness of serotonin precursors in altering mood states. A Japanese study supplemented 100 patients with either 5-HTP or placebo and observed a significant improvement in mood in 69% of treated patients after two weeks [source]. A follow up study in Switzerland compared the effectiveness of 5-HTP supplementation to SSRIs in patients with poor mood. In a double-blind, multicenter study, both treatment groups showed near equal reduction in depressive characteristics by the end of six weeks[source]. The effective delivery of serotonin precursors and their efficacy remains a topic of investigation, yet the present clinical data that exists suggests 5-HTP and L-tryptophan supplementation can aid in promoting positive mood.

  3. The ingredients in this product are far safer than the chemicals in most antidepressant and antianxiety medications. I take most of these ingredients individually, and have NONE of the really bothersome side effects I was getting with Zoloft, Welbutrin, Celexa, OR Effexor. I may give these a try!
